Abstract:Objective To investigate the effects of IL-27 gene on phenotype and function of DCs (Dendritic cell) derived from PBMCs (peripheral blood mononuclear cells). Methods isolated from healthy human peripheral blood were cultured with GM-CSF (granulocyte-macrophage clone stimulating factor)plus IL(Interleukin)-4 for 5 days, and then were divided into three groups: IL-27 gene transfection, TNF (tumor necrosis factor)-α and control group. The morphological characteristics of DCs were observed by inverted microscope. Expressions of CD1a, CD83 and CD80, and CD86 on DCs were analyzed with flow cytometry.MTT method was used to determine the proliferation of T cells stimulated by DCs.ELISA was used to detect the production of IL-12 and IFN-γ in the culture supernatant of DCs. Results PBMCs were developed into mature DCs with typical morphological characteristics after IL-27 gene transfection.Expressions of CD1a,CD83 and CD80,and CD86 on DCs in IL-27 gene transfection and TNF-α groups were up-regulated compared to the control group (
P<0.05).IL-27 gene transfected DCs markedly stimulated the proliferation of T cells and enhanced the production of IL-12 and IFN-γ (
P<0.05). Conclusion IL-27 gene transfection could up-regulate the phenotype of mature DCs and enhance the immune activity of DCs.
